Dukino's mom is an enormous skag (an even bigger skag than Skagzilla from Borderlands 1) found in Lynchwood. You are required to kill her in the quest titled "Demon Hunter", if you have completed the necessary requirements. In order to unlock the mission, you must help her kid Dukino, a sick baby skag found at the end of the main town in Lynchwood.
An easy way to defeat her is to simply remain in the elevator you used to descend into the cave. From there, you will be able to easily take her out without taking much, or any, damage. Her attacks include melee at close range (not an issue if you remain in the lift), lobbed electric spheres, and a "death" ray. If you remain in the lift, just be aware of her death ray, and duck behind the railing as she sweeps it across the screen.
Dukino's mom has thick armor, so it is recommended that you use a corrosive weapon. During the fight, many low level bandits will also try to kill the gigantic skag, but will immediately be annihilated. Once she is defeated, complete the mission by finding Dukino in the center of the cave, which allows you to turn in. You will be rewarded with the Buffalo rifle.
